Centre's James Kusak scored on Cameron Bayse's assist at 36:38 for the game's only goal.
The Colonels outshot the Tigers 13-8, while DePauw held a 4-3 advantage in corners. DePauw's Ted Jacobi recorded three saves in goal, while Centre's Aaron Arndt totaled four.
DePauw travels to Rose-Hulman on Wednesday for an 8 p.m. contest.
