Allison Baccich and Carolyn Mueller scored first-goals before Melissa Davis and Mueller each scored in the second half. St. Vincent's Ginger Paddock scored with 1:20 remaining.
DePauw outshot St. Vincent 19-6 and held an 18-2 advantage in corners.
The Tigers travel to Bellarmine on Saturday.
